Mechanism of Action

  • The hypolipidemic activity of Guggulsterones could be attributed to several mechanisms including inhibition of cholesterol biosynthesis and enhancement in cholesterol degradation and/ or excretion. 
  • Guggulsterones E and Z act as antagonists at the farnesoid X receptor, allowing more cholesterol catabolism and excretion from the body. This receptor mediates the conversion of cholesterol to bile acids; by antagonizing this receptor, 7α-hydroxylase is released, stimulating cholesterol catabolism. This helps to maintain the cholesterol and bile acid homeostasis accounting for itshypolipidemic activity.
  • Another mechanism proposed is that Guggulsterone upregulates the expression of the bile salt export pump (BSEP), a rate-limiting efflux transporter for eliminating cholesterol metabolites bile acids from the liver. Such upregulation is possibly mediated through the activating protein 1 (AP-1) signaling pathway The FXR antagonism and enhanced BSEP expression have been proposed as possible mechanisms for the hypolipidemic effect of guggulsterone
  • Guggul markedly inhibits liver cholesterol biosynthesis. This causes interference in lipoprotein formation and lipid turnover. 
  • Guggulincreases fecal excretion of bile acids (cholic and deoxycholic acids) & cholesterol and lowers intestinal absorption of fat and cholesterol. Guggul stimulates the LDL receptor binding activity in hepatocytes and enhances its catabolism.Guggulsterones have the capability of inhibitingoxidative modification of LDL.
